This is an all-inclusive cell-based luciferase reporter assay kit targeting the Dog Liver X Receptor Beta (dLXR Beta). INDIGO’s dLXR Beta reporter assay utilizes proprietary mammalian cells that have been engineered to provide constitutive expression of the dLXR Beta. In addition to dLXR Beta Reporter Cells, this kit provides two optimized media for use during cell culture and in diluting the user’s test samples, a reference agonist, Luciferase Detection Reagent, and a cell culture-ready assay plate. The principal application of this assay is in the screening of test samples to quantify any functional activity, either agonist or antagonist, that they may exert against dLXR Beta. This kit provides researchers with clear, reproducible results, exceptional cell viability post-thaw, and consistent results lot to lot. Kits must be stored at -80C. Do not store in liquid nitrogen. Note: reporter cells cannot be refrozen or maintained in extended culture.

Dog LXRb agonist dose-response assays. Dog LXRb Reporter Cells were treated with the reference agonists T0901317 (provided) and GW3965 (Tocris). Luminescence was quantified and values of Average RLU and corresponding standard deviation and percent coefficient of variation, and Fold-Activation were calculated for each treatment group (n = 4). Z’ values were calculated as described by Zhang, et al. (1999). GraphPad Prism software was used to perform the least-squares method of non-linear regression to curve-fit data and determine EC50 values.

Target Background

Liver X receptor β (LXRβ) is a member of the nuclear receptor family of transcription factors. LXRβ is encoded by the NR1H2 gene (nuclear receptor subfamily 1, group H, member 2). The Liver X receptors (LXRs) were originally identified as orphan members of the nuclear receptor superfamily because their ligands were unknown. Like other receptors in the family, LXRs heterodimerize with retinoid X receptor and bind to specific response elements (LXREs) characterized by direct repeats separated by 4 nucleotides.
