Nuclear Receptor Assays
INDIGO’s Nuclear Receptor Assays are reporter assays for intracellular transcription factors that directly regulate gene expression in response to lipophilic molecules commonly referred to as nuclear receptors (NRs). Nuclear receptors affect a wide variety of functions, including fatty acid metabolism, reproductive development, and detoxification of foreign substances. Many NRs act as ligand-inducible transcription factors, responding to endogenous and exogenous chemicals. Due to this, nuclear receptors are important therapeutic targets in the behavior of many diseases including autoimmune/inflammation, cardiovascular disease, cancer, NASH, and many others.
INDIGO’s nuclear receptor assays utilize bio-luminescence reporter gene technology. Bioluminescence technologies such as firefly luciferase reporter assay kits provide researchers with increased sensitivity and precision due to low signal-to-noise ratio. INDIGO’s reporter cells incorporate the firefly luciferase reporter gene functionally linked to a nuclear receptor specific promoter. Following ligand-activation, the nuclear receptor complex acts to induce expression of the luciferase reporter. Upon addition of detection reagent, the intensity of light emission from the luciferase reaction directly correlates to the activation status of the specific nuclear receptor. Luminescence intensity of the reaction is quantified using a luminometer and is reported in terms of Relative Light Units (RLU’s). Thus, quantifying changes in luciferase expression in the test sample-treated reporter cells provides a sensitive surrogate measure of changes in activity.
INDIGO’s nuclear receptor assays give researchers the ability to screen compounds to quantify agonist or antagonist activities against specific nuclear receptors expressed within the cytoplasm and nuclear environments of healthy, dividing cells. These nuclear receptor assays can be utilized for high throughput screening against one target, to understand a compounds potency and develop EC50/IC50 values, as well as to profile compounds against a panel of nuclear receptor assays to evaluate for selectivity.

INDIGO's nuclear receptor assay kits provide the convenience of an all-inclusive cell-based assay system. In addition to nuclear receptor specific reporter cells, our kits include two optimized media for use in recovering the cryopreserved cells and for diluting test samples, the reference agonist or inhibitor of the nuclear receptor reporter system, Luciferase Detection Reagents, and a cell culture-ready assay plate.
Nuclear Receptor Specific Reporter Cells are transiently transfected and prepared as frozen stocks using INDIGO's proprietary CryoMite™ process. This cryo-preservation method allows for the immediate dispensing of healthy, division-competent reporter cells into assay plates. There is no need for cumbersome intermediate treatment steps such as spin-and-rinse of cells, viability determinations, or cell titer adjustments prior to assay setup.
