INDIGO’s Thyroid Hormone Receptor Panel (TR alpha, TR beta) assay kit is an all-inclusive firefly luciferase reporter assay system that includes addition to TR Reporter Cells, two optimized media for use during cell culture and (optionally) in diluting the user’s test samples, a reference agonist, Luciferase Detection Reagent, a cell culture-ready assay plate, and a detailed protocol. This panel contains sufficient materials to perform 48 TR alpha assays and 48 TR beta assays in 96-well plate format.

Target Background
INDIGO’s PANEL of TR Thyroid Hormone Receptor Panel Reporter Assays utilizes non-human mammalian cells engineered to express Human Thyroid Hormone Receptors Alpha (NR1A1) and Beta (NR1A2), ligand-dependent transcription factors commonly referred to as TRα and TRβ. Because each of these reporter cells incorporates a responsive luciferase reporter gene, quantifying expressed luciferase activity provides a sensitive surrogate measure of TR activities in the treated cells.
The principle application of this reporter assay system is in the screening of test samples to quantify any functional activity, either agonist or antagonist, that they may exert against human TRα and TRβ.
