This is an all-inclusive cell-based luciferase reporter assay kit targeting the Mouse Liver X Receptor Alpha (mLXRa). INDIGO’s mLXRa reporter assay utilizes proprietary mammalian cells that have been engineered to provide constitutive expression of the mLXRa. In addition to mLXRa Reporter Cells, this kit provides two optimized media for use during cell culture and in diluting the user’s test samples, a reference agonist, Luciferase Detection Reagent, and a cell culture-ready assay plate. The principal application of this assay is in the screening of test samples to quantify any functional activity, either agonist or antagonist, that they may exert against mLXRa. This kit provides researchers with clear, reproducible results, exceptional cell viability post-thaw, and consistent results lot to lot. Kits must be stored at -80C. Do not store in liquid nitrogen. Note: reporter cells cannot be refrozen or maintained in extended culture.

Mouse LXRa dose-response assays. Mouse LXRa Reporter Cells were treated with the reference agonist TO901317. Luminescence per assay well was quantified and Average RLU, standard deviation, percent coefficient of variation, and Average Fold-Activation were calculated for each treatment group (n=4). Z’ values were calculated as described by Zhang, et al. (1999). Non-linear regression and EC50 analyses were performed using GraphPad Prism software.

Target Background

Liver X receptor α (LXR-α or LXRα) is a nuclear receptor protein encoded by the NR1H3 gene (nuclear receptor subfamily 1, group H, member 3). The Liver X receptors (LXRs) were originally identified as orphan members of the nuclear receptor superfamily because their ligands were unknown. Like other receptors in the family, LXRs heterodimerize with retinoid X receptor and bind to specific response elements (LXREs) characterized by direct repeats separated by 4 nucleotides.
